Bei einem Klassentreffen gelingt dem früheren Bösewicht Gru ein echter Fang. Im Auftrag der Anti-Verbrecher-Liga verhaftet er Ex-Mitschüler Maxime Le Mal, mit dem er schon in der Schulzeit verfeindet war. Als dieser nur wenig später aus dem Gefängnis entkommt und auf Rache sinnt, müssen Gru und seine Frau Lucy mit ihrem vor kurzem geborenen Baby Gru Junior und ihren Adoptivtöchtern Margo, Edith und Agnes im beschaulichen Mayflower untertauchen. Nicht aufzufallen ist jedoch ganz schön schwer.
